Comment | The transcription start site was deduced from the sequences conserved between the human, rat, and mouse IFN-gamma genes reported by Penix et al., J. Exp. Med., 178, 1483-1496 (1993). The first exon of the mouse IFN-gamma gene has been revised in NCBI Reference Sequence: NM_008337.4. In pGL4_ifng-2500-luc, the PCR product (2.6 kb) was amplified with forward primer (5' CTA CTC GAG AAA TGG GAC ACA GAT CTC 3') and reverse primer (5' CTA GAT ATC TGT CTC AGA GCT AGG CCG CAG GAG GAG AAG C 3'), digested with Xho I and EcoRV, and inserted into the Xho I and EcoRV sites of pGL4.22[luc2CP/Puro] vector. Genomic DNA fragment 118,438,542 to 118,441,154 nt of NC_000076.6 is cloned. Known differences (at least): C deletion at 118,438,965 nt; A insertion between 118,440,965 and 118,440,966 nt. |
