Strain development | Developed by Arikuni Uchimura and Takeshi Yagi, Graduate School of Frontier Biosciences, Osaka University in 2007. C57BL/6J derived ES cells were used to generate the knockout mice. The polyA-neo cassette was removed by crossing with Sycp1-cre mice. The mutant mice were crossed to C57BL/6J. |
Strain description | Pold1 (polymerase (DNA directed) D400A variant mutant mice lacking 3’-5’exonuclease activity in the Pol delta. About 30% of homozygous mutant mice die with abnormally swollen thymus. Most surviving mice develop nodules on their tails. It is preferable to avoid a mating using heterozygous male across continuous generations (2 generations and up) to avoid an increasing mutation rate. |
