Appearance
Strain developmentBRUCE-4 ES cell line (derived from C57BL/6J mouse line) was used for homologous recombination. The mice were generated by Dr. Tomohiko Okuda et al. in Aburahi Laboratories of SHIONOGI & CO., LTD. in 2010. Chimeras generated using these ES cells were bred with C57BL/6JJcl (CLEA Japan, Inc.). After confirmation of germ-line transmission, homozygous knockout mice were generated by inbreeding of heterozygous mice. B6-Mboat4tm1Shi homozygotes were maintained via incrossing. The engineered mutation was transmitted to the progeny with the expected Mendelian ratio and develop normally.
Strain descriptionA null KO mouse of the Mboat4 gene encoding ghrelin O-acyltransferase (GOAT). Exon1 of the gene was replaced with a PGK-Neo cassette by homologous recombination in ES cells. Homozygous KO mice show no acylated ghrelin. This model mouse can be utilised for reserches of diseases, inter alia, metabolic syndrome.
