Strain developmentFucci2 consists of the cell cycle phase specific biosensors mCherry-Cdt1(30/120) and mVenus-hGem(1/110) fused with the Thosea asigna virus 2A peptide sequence in a single bicistronic transgene (Mort et al., 2014).Arl13bCerulean consists of the full mouse Arl13b cDNA (OTTMUST00000058920.GRCm38) and mCerulean. Arl13bCerulean-Fucci2a couples Arl13bCerulean to Fucci2a using the porcine teschovirus-1 2A (P2A) peptide on the N-terminus of Fucci2a to create a tricistronic construct. The CAG promoter incorporates the cytomegalovirus (CMV) early enhancer; the promoter, first exon and intron of the chick β-actin gene; and the beta-globin splice acceptor (Miyazaki et al., 1989).
