Strain development | Dr. Jun Aruga, RIKEN Brain Science Institute (2005). Mice with germline transmission were first mated with mice transgenic for Cre recombinase under the control of the cytomegalovirus immediate early enhancerchicken b-actin hybrid (CAG) promotor. Mice carrying the mutated Slitrk2 allele were backcrossed to C57BL/6J for more than eight generations. Slitrk2 knockout (KO) mice exhibited increased locomotor activity in novel environments, antidepressant-like behaviors, enhanced vestibular function, and increased plasticity at mossy fiber–CA3 synapses with reduced sensitivity to serotonin. A serotonin metabolite was increased in the hippocampus and amygdala, and serotonergic neurons in the raphe nuclei were decreased in Slitrk2 KO mice. When Slitrk2 KO mice were treated with methylphenidate, lithium, or fluoxetine, the mood stabilizer lithium showed a genotype-dependent effect. Slitrk2 KO mice are useful for studies of neuropsychiatric disorders. |
Strain description | Mutation details: A targeting vector was designed to delete a 5.8 kb region containing entire open-reading frame. A floxed neo replaced the sequence and was subsequently excised by crossing with mice expressing Cre in the germ line. Western blot of mutant brain samples showed no bands corresponding to Slitrk2 protein. |
