Strain development | 1. The Iqgap3 gene (NCBI Reference Sequence: NM_001033484.1) is located on mouse chromosome 3.Thirty-eight exons have been identified, with the ATG start codon in exon 1 and the TGA stop codon in exon 38.2. In the targeting vector, the TGA stop codon will be replaced with the “2A-tdTomato” cassette.3. To engineer the targeting vector, homology arms will be generated by PCR using BAC cloneRP24-71K14 or RP23-68H24 from the C57BL/6 library as template.4. In the targeting vector, the Neor cassette will be flanked by SDA (self-deletion anchor) sites. DTA will be used for negative selection.5. C57BL/6 ES cells will be used for gene targeting.1. Targeted C57BL/6 ES cell clone was injected into C57BL/6 albino embryos, which were then re-implanted into CD-1 pseudo-pregnant females.2. Founder animals were identified by their coat color, their germline transmission was confirmed by breeding with C57BL/6 females and subsequent genotyping of the offspring.3. Heterozygous targeted mice were generated and maintain strain by crossing with wild type C57BL/6 mice. |
Strain description | Iqgap3 is a cytosolic scaffold protein mainly expressed in stem/progenitor cells. Iqgap3-tdTomato mouse strain can be used to monitor the expression pattern of Iqgap3 by red fluorescent protein tdTomato.Endogenous Iqgap3 expression is intact. The mice do not show abnormality, and the female mice can get pregnant normally. |
