Strain development | 1. The Runx3 exon2-4 was isolated from C57BL/6 BAC clone using high-fidelity polymerase chain reaction.2. PCR-based Site-direct mutagenesis was performed to introduce the point mutation R122C in exon 3.3. The targeting vector was generated by cloning the genomic fragments containing the intronic region between exon 3 into the polylinker of the target vector NEO-DTA cassette contained a Pgk-Neor (neomycin resistant) gene flanked by LoxP sites and a DTA (diphtheria toxin) cassette for the selection.4. TT2 ES cells will be used for gene targeting.5. The neomycin expression cassette was then excised by crossing the chimera mice with EIIa-Cre mice. (Douchi et al., CMGH 2022)1. Targeted TT2 ES cell clone which has R123C mutation (equivalent of human RUNX3 R122C mutation) in exon 3 of Runx3 gene was injected into C57BL/6 embryos, which were then re-implanted into pseudo-pregnant females.2. Germline transmission was confirmed by genotyping of the offspring.3. Heterozygous targeted mice were generated and maintain strain by crossing with wild type C57BL/6 mice.4. This line was backcrossed to C57BL/6 for more than ten generations by the donating laboratory.5. This line is maintained as homozygote. |
Strain description | Runx3 is a transcription factor and functions as a developmental regulator. Runx3 is frequently associated with cancer. R122C mutation (equivalent of mouse Runx3 R123C mutation) was originally identified in human gastric cancer. 30 % of Runx3-R122C/R122C homozygote mice show some precancerous metaplasia at 6-month age. The homozygote female mice can get pregnant normally. The homozygote mice can survive more than one year. |
