Strain development | 1. The Runx3 gene (NCBI Reference Sequence: NM_019732.2) is located on mouse chromosome 4. Six exons have been identified, with the ATG start codon in exon 1 and TAA stop codon in exon 6.2. For the KI model, the “loxP-Endogenous SA of intron 2-Runx3 exon 3~6 CDS-ployA-SDA-Neo-SDA-loxP” cassette was inserted into intron 2, the p.R137C (CGC to TGC) was introduced into exon 3 in 3’ homology arm.3. In the targeting vector, the Neo cassette was flanked by SDA (self-deletion anchor) sites. DTA will be used for negative selection.4. C57BL/6 ES cells will be used for gene targeting.Mouse Runx3 conditional knockin model in C57BL/6N mice.1. Targeted C57BL/6N ES cell clone was injected into C57BL/6 albino embryos, which were then re-implanted into CD-1 pseudo-pregnant females.2. Founder animals were identified by their coat color, their germline transmission was confirmed by breeding with C57BL/6N females and subsequent genotyping of the offspring.3. Heterozygous targeted mice were generated, and strains maintained by crossing with wild type C57BL/6N mice. |
Strain description | Runx3 is a transcription factor and functions as a developmental regulator. Runx3 is frequently associated with cancer. R122C mutation was identified in human gastric cancer. LSL-Runx3 R122C mouse strain expresses wild-type Runx3, which is regulated by endogenous Runx3 promoter activity, and the strain does not show any phenotype. By crossing with promoter-specific Cre mouse, wild-type Runx3 expression will be replaced with the mouse equivalent of R122C in Cre-expressing cells (i.e. R123C in mouse, henceforth referred as R122C for clarity and historical reasons). The homozygote female mice can get pregnant normally. The homozygote mice can survive more than one year. |
